目的:探寻大孔树脂分离精制复方三子咳喘胶囊浸膏的最佳工艺条件。方法:通过对3种不同类型的大孔吸附树脂吸附洗脱特性参数比上柱量(Saturation ratio)、比吸附量(Absorbtion ratio)和比洗脱量(Eluation ratio)的比较研究确定大孔树脂的类型和洗脱溶剂。随后对影响吸附分离的主要因素如上柱药液浓度、上柱药液量、上柱药液温度进行正交试验筛选,并结合浸膏得率和芥子碱硫氰酸盐含量两个指标进行正交评价。结果:选择吸附参数最大的天津南开大学生产的AB-8型大孔树脂和70%浓度的乙醇进行分离洗脱,将浓缩至2:1(生药量:体积)的温度为20℃的复方溶液以1.5:1的比例(溶液体积:树脂量)进行上柱分离洗脱为最佳条件。结论:AB-8型大孔树脂适宜三子咳喘胶囊浸膏的分离精制。
