目的:以人肾癌786-O细胞作为体外抗肿瘤实验模型,筛选吉祥草的抗肿瘤活性部位。方法:采用MTT法筛选乙醇提取物及其不同溶剂萃取部分对786-O细胞体外抗肿瘤作用的活性部位;通过Hoechst 33258染色于荧光显微镜下观察细胞凋亡形态;应用流式细胞术AnnexinⅤ-FITC/PI双标记法检测细胞凋亡率。结果:吉祥草正丁醇萃取物对786-O细胞的抑制作用较强且呈量效和时效关系,作用48 h时IC50值为69.33 mg/L;凋亡染色显示有典型的凋亡形态出现;流式细胞仪检测吉祥草正丁醇萃取物能诱导786-O细胞凋亡并呈剂量依赖关系。结论:正丁醇部位是吉祥草抗肿瘤作用的主要活性部位,吉祥草正丁醇萃取物能抑制人肾癌786-O细胞增殖并诱导细胞凋亡。
OBJECTIVE: To screen the anti-tumor active site of auspicious grass by using human renal carcinoma 786-O cells as an in vitro anti-tumor experimental model. METHODS: MTT was used to screen the active site of antitumor effect of ethanol extract and its different solvent extraction on 786-O cells in vitro. The morphology of apoptotic cells was observed under fluorescence microscope with Hoechst 33258 staining. The apoptosis of 786-O cells was detected by flow cytometry Annexin V-FITC / PI double labeling method to detect apoptosis rate. Results: The extract of Jiuxiancao n-butanol had stronger inhibitory effect on the 786-O cells in a dose-effect and time-dependent manner, with an IC50 value of 69.33 mg / L at 48 h. Apoptotic staining showed typical apoptotic appearance Flow cytometry detection of n-butanol extracts of Ji Xiang Cao induced 786-O cell apoptosis in a dose-dependent manner. CONCLUSION: The n-butanol fraction is the main active site of antitumor activity of Ji auspicious grass. Butanol extract of auspicious grass can inhibit the proliferation of human renal carcinoma 786-O cells and induce apoptosis.
