目的 :研究黄酮类化合物槲皮素对白血病细胞株 NB4的抑制作用。方法 :应用细胞计数、细胞形态、DNA凝胶电泳等方法 ,在体外研究槲皮素诱导 NB4细胞凋亡的作用。结果 :10μmol/ L浓度的槲皮素无明显抑制 NB4增殖作用 ,过高的浓度可能发生细胞变性 ,而当作用浓度为 30μm ol/ L~ 90μmol/ L时 ,就能显示抑制效应 ,其抑制作用呈剂量依赖性 ,细胞表现典型的凋亡特征。结论 :槲皮素在一定的浓度范围内能抑制 NB4细胞增生 ,其作用随时间和浓度增加而增强 ,并诱导凋亡。
Objective: To study the inhibitory effect of flavonoid quercetin on leukemia cell line NB4. Methods: The effects of quercetin on the apoptosis of NB4 cells were studied in vitro by cell counting, cell morphology and DNA gel electrophoresis. Results: Quercetin at a concentration of 10 μmol / L did not significantly inhibit the proliferation of NB4 cells, and cell degeneration might occur at high concentration. When the concentration was 30 μmol / L ~ 90 μmol / L, the inhibitory effect was inhibited In a dose-dependent manner, the cells showed typical apoptotic features. CONCLUSION: Quercetin inhibits the proliferation of NB4 cells in a certain concentration range. The effect of quercetin increases with time and concentration, and induces apoptosis.
