本研究旨在探讨活化的T细胞对间充质干细胞(mesenchymal stem cell,MSC)向成骨细胞分化的影响。将活化的T细胞与MSC共培养,14天后碱性磷酸酶染色,观察成骨细胞的生成。此外,在MSC的培养体系中加入活化的T细胞上清,RT-PCR检测活化的T细胞免疫相关分子的表达,探讨哪些细胞因子参与了MSC向成骨细胞分化。结果表明,活化的T细胞能促进MSC分化为成骨细胞,IL-1β在其中发挥重要的作用,而TNF-α,TGF-β1作用不明显。结论:活化的T细胞促进MSC向成骨细胞分化,MSC与免疫细胞通过细胞因子发生相互作用。
This study aimed to investigate the effect of activated T cells on osteoblastic differentiation of mesenchymal stem cells (MSCs). Activated T cells were co-cultured with MSCs, stained with alkaline phosphatase 14 days later, and osteoblast formation was observed. In addition, activated T cell supernatants were added to the culture system of MSC. The expression of immune-related T cells was detected by RT-PCR and the cytokines involved in MSC differentiation into osteoblasts. The results showed that activated T cells can promote the differentiation of MSC into osteoblasts, IL-1β play an important role in them, while the effect of TNF-α and TGF-β1 is not obvious. CONCLUSION: Activated T cells promote MSC differentiation into osteoblasts, and MSC interact with immune cells through cytokines.
