采用组织块法从皖西大别山野生博落回Macleaya cordata中分离内生真菌,根据形态学特征对其进行类型归类,采用薄层色谱(TLC)和高效液相色谱(HPLC)鉴定其内生真菌次级代谢产物,运用形态学方法对活性菌株进行初步菌种鉴定。实验结果显示,分别从博落回根、茎、叶中分离出28,57,96株内生真菌。根据形态学特征,初步把其归类为26个类型。其中菌株BLH 51的发酵液中含有血根碱,经形态学初步鉴定为层出镰刀菌Fusarium proliferatum。表明药用植物博落回组织中含有较丰富的内生真菌,可以成为寻找生物活性代谢产物的一个新来源。
Endophytic fungi were isolated from the wild Macleaya cordata in the Dabieshan Mountains of western Anhui using tissue-block method. The endophytic fungi were classified according to morphological characteristics and identified by TLC and HPLC Fungal secondary metabolites, the use of morphological methods for the initial identification of strains of bacteria. The experimental results showed that 28, 57, 96 endophytic fungi were isolated from the roots, stems and leaves of Plutella xylostella respectively. According to morphological characteristics, initially classified as 26 types. Among them, the fermentation broth of strain BLH 51 contained sanguinarum and was initially identified as Fusarium proliferatum by morphology. The results showed that the medicinal plant Botanica came back to the tissue rich in endophytic fungi, which could become a new source for searching bioactive metabolites.
