以野生仙人掌为材料,通过单因素试验及L9(34)正交试验对仙人掌多糖的热水浸提工艺进行优化;优化工艺条件下,比较茎龄为2~4个月及茎龄大于5个月的仙人掌多糖提取结果;并通过对羟基自由基(·OH)的清除比较不同提取方法得到仙人掌多糖的体外抗氧化活性。结果表明,最佳提取工艺条件为:液料比60︰1(mL/g)、提取时间3 h、提取温度95℃,此优化条件下茎龄为2~4个月的野生仙人掌多糖提取率为30.28%±0.22%,极显著高于茎龄大于5个月的野生仙人掌多糖提取率(13.85%±0.08%);2种多糖纯度及蛋白含量无显著性差异。水提仙人掌多糖有明显的抗氧化活性,其中多糖质量浓度为7.5 mg/mL时,对·OH的清除率为57.7%,与2 mg/mL VC的·OH清除率56.0%相当。
Using wild cactus as material, the hot water extraction process of cactus polysaccharide was optimized by single factor test and L9 (34) orthogonal test. There was no significant difference in purity and protein content between the two polysaccharides. Water extract cactus polysaccharide has obvious antioxidant activity, of which polysaccharide concentration of 7.5 mg / mL, the clearance rate of · OH was 57.7%, and 2 mg / mL VC · OH clearance rate of 56.0%.
