目的:探讨产妇子宫平滑肌大电导钙激活钾通道(BKCa)与小窝蛋白相互作用在宫缩乏力性产后出血发病中的作用。方法:采用免疫荧光双染法及激光共聚焦显微镜观察30例宫缩乏力性产后出血产妇(病例组)和30例宫缩正常无产后出血产妇(对照组)子宫平滑肌BKCa(α亚基)与小窝蛋白-1、BKCa(α亚基)与小窝蛋白-2的共定位情况;采用免疫共沉淀法检测两组产妇子宫平滑肌BKCa(α亚基)与小窝蛋白-1、BKCa(α亚基)与小窝蛋白-2的相互作用关系。结果:(1)两组子宫平滑肌细胞均有BKCa(α亚基)、小窝蛋白-1和小窝蛋白-2表达,其中BKCa(α亚基)主要位于细胞膜表面,而小窝蛋白-1和小窝蛋白-2在细胞膜表面和细胞质均有表达;且BKCa(α亚基)与小窝蛋白-1、BKCa(α亚基)与小窝蛋白-2均有部分区域共表达于细胞膜。(2)病例组子宫平滑肌BK-Ca(α亚基)免疫沉淀物中小窝蛋白-1、小窝蛋白-2蛋白水平均高于对照组,差异均有统计学意义(P<0.05)。结论:宫缩乏力性产后出血产妇子宫平滑肌细胞中BKCa(α亚基)免疫沉淀物中小窝蛋白-1、小窝蛋白-2蛋白水平升高,提示小窝蛋白与BKCa(α亚基)相互作用增强可能是宫缩乏力性产后出血发病的重要原因之一。
Objective: To investigate the role of the interaction between BKCa and caveolin in uterine smooth muscle in the development of uterine inert postpartum hemorrhage. Methods: Thirty cases of uterine smooth muscle BKCa (α subunit) and 30 cases of normal uterine bleeding without maternal uterine bleeding (control group) were observed by immunofluorescence double staining and confocal laser scanning microscope The co-localization of caveolin-1 and BKCa (α subunit) and caveolin-2 was detected by immunoprecipitation. The expressions of BKCa (α subunit) and caveolin- Subunit) and caveolin-2 interaction. Results: (1) BKCa (α subunit), caveolin-1 and caveolin-2 were expressed in both groups of uterine smooth muscle cells. BKCa (α subunit) was mainly located on the surface of cell membrane, And caveolin-2 were expressed on the cell membrane surface and in the cytoplasm; and some regions of BKCa (α subunit) and caveolin-1, BKCa (α subunit) and caveolin-2 were co-expressed in the cell membrane. (2) The protein expressions of caveolin-1 and caveolin-2 in BK-Ca (α subunit) immunoprecipitates of uterine smooth muscle were higher than those in control group (P <0.05). Conclusions: The protein expressions of caveolin-1 and caveolin-2 in BKCa (α subunit) immunoprecipitates in uterine smooth muscle cells of uterine bleeding postpartum hemorrhage are both higher than those in BKCa (α subunit) Interaction enhancement may be one of the important causes of uterine atony postpartum hemorrhage.
