目的探讨发酵冬虫夏草菌粉对糖尿病大鼠肾脏细胞凋亡及B细胞淋巴瘤/白血病2(Bcl-2)、Bcl-2相关X蛋白(Bax)表达的影响。方法 24只SD大鼠均分为糖尿病(A)组、治疗(B)组和对照(C)组,A、B组饲喂高糖高脂饲料8周并注射链脲佐菌素30mg/kg建立糖尿病大鼠模型,C组饲喂普通饲料作对照。B组成模后予发酵冬虫夏草菌粉200mg·kg-1·d-1灌胃8周,A、C组予等体积无菌水灌胃。计算三组内生肌酐清除率(CCr)、24-h尿白蛋白排泄率(UAER),流式细胞术检测肾脏细胞凋亡率,免疫组化法检测肾组织Bcl-2和Bax蛋白表达。结果与C组相比,A、B组CCr降低,24-h UAER、肾脏细胞凋亡率和肾组织Bcl-2和Bax表达升高(P<0.05或P<0.01)。与A组相比,B组CCr和肾小管上皮细胞Bcl-2表达升高,24-h UAER、肾脏细胞凋亡率和肾小管上皮细胞Bax表达降低(P<0.05)。结论发酵冬虫夏草菌粉能抑制糖尿病大鼠肾脏细胞凋亡,与影响Bcl-2/Bax基因表达有关。
Objective To investigate the effects of fermented Cordyceps sinensis on renal cell apoptosis and the expression of Bcl-2 and Bcl-2 in kidney of diabetic rats. Methods 24 SD rats were divided into diabetic group (A), treatment group (B) and control group (C). Groups A and B were fed with high glucose and high fat diet for 8 weeks and received streptozotocin 30 mg / kg The diabetic rat model was established, and the C group was fed with normal diet as control. Group B was fed with 200 mg · kg -1 · d -1 of fermented Cordyceps sinensis powder for 8 weeks after oral administration. Groups A and C received equal volume of sterile water. Three groups of endogenous creatinine clearance rate (CCr), 24-hour urinary albumin excretion rate (UAER) were calculated. Apoptosis rate of renal cells was detected by flow cytometry. Bcl-2 and Bax protein expression was detected by immunohistochemistry. Results Compared with group C, the CCr in group A and group B were decreased, the rate of 24-h UAER, the rate of renal cell apoptosis and the expression of Bcl-2 and Bax in renal tissue were increased (P <0.05 or P <0.01). Compared with group A, the expression of Bcl-2 in CCr and tubular epithelial cells in group B was increased, while the apoptosis rate in 24-h UAER, renal cell and tubular epithelial cells was decreased (P <0.05). Conclusion Fermentation of Cordyceps Sinensis powder can inhibit renal cell apoptosis in diabetic rats, which may be related to the effect of Bcl-2 / Bax gene expression.
