目的:观察动脉粥样硬化(AS)患者血管中胶原含量和组织蛋白酶B的表达水平,探讨其对AS的作用。方法:9例AS患者(AS组)和5例正常成人(对照组)动脉血管标本,采用Van Gieson法染色检测胶原含量,应用免疫组化染色测定细胞浆内平均积分吸光度值,运用Western Blotting法检测组织蛋白酶B的光密度比值。结果:Van Gieson法染色观察到AS组胶原不同程度断裂、分布不均,并有大量炎性细胞浸润;对照组血管壁胶原分布均匀,排列整齐,无明显炎症细胞浸润。与对照组胶原含量(5.10±1.10%)相比,AS组(7.80±1.60%)明显升高(P<0.05)。
Objective: To observe the content of collagen and the expression of cathepsin B in the blood vessels of patients with atherosclerosis (AS) and to explore its effect on AS. Methods: Arterial blood vessels of 9 AS patients (AS group) and 5 normal adult (control group) arteries were collected. The content of collagen was detected by Van Gieson staining. The average integral absorbance of cytoplasm was measured by immunohistochemical staining. The optical density ratio of cathepsin B was measured. RESULTS: Van Gieson staining showed that the collagen in the AS group was broken, unevenly distributed and infiltrated with a large number of inflammatory cells. The collagen in the vascular wall of the control group was evenly distributed and arranged neatly with no obvious inflammatory cell infiltration. Compared with control group (5.10 ± 1.10%), AS group (7.80 ± 1.60%) was significantly increased (P <0.05).
