目的通过体外细胞培养实验探讨茶多酚对人肺癌细胞H460生长抑制作用及其作用机制。方法体外培养人肺癌H460细胞,分对照组及茶多酚药物组(茶多酚终浓度为100μg/ml),加药后继续培养12h、24h、36h,应用荧光显微镜下观察细胞核的形态变化,流式细胞仪观察茶多酚诱导细胞凋亡的作用及阻滞细胞周期的作用。结果 MTT法显示茶多酚可抑制肺癌细胞的增殖活性,其抑制作用随时间的延长而增强。流式细胞术发现茶多酚可使肺癌细胞阻滞于G1期。
Objective To investigate the inhibitory effect of tea polyphenols on the growth of human lung cancer cell line H460 in vitro and its possible mechanism. Methods Human lung cancer H460 cells were cultured in vitro and divided into the control group and the tea polyphenols group (the final concentration of tea polyphenols was 100 μg / ml). The cultured cells were cultured for 12h, 24h and 36h. The morphological changes of the cells were observed under a fluorescence microscope. Flow cytometry observation of tea polyphenols induced apoptosis and the role of cell cycle arrest. Results MTT method showed that tea polyphenols can inhibit the proliferation activity of lung cancer cells, and its inhibitory effect increased with time. Flow cytometry found that polyphenols in lung cancer cells can be arrested in the G1 phase.
