目的:优选复方通脉颗粒的大孔树脂纯化工艺。方法:以总酚酸,葛根素的吸附-洗脱率为指标,采用单因素试验考察上样液质量浓度、径高比、洗脱剂等对复方通脉颗粒大孔树脂纯化工艺的影响。结果:优选的纯化工艺为上样液质量浓度110 g.L-1,树脂柱径高比1∶10,树脂体积与上样量比2.8∶1(吸附3 h),加2BV水洗除杂,用6 BV 50%乙醇洗脱,收集洗脱液。结论:该纯化工艺合理、稳定,可推广于大生产应用。
Objective: To optimize the macroporous resin purification process of Fufang Tongmai granules. Methods: The adsorption and elution rates of total phenolic acids and puerarin were used as indexes. The effects of the concentration of the sample solution, the aspect ratio and the eluent on the purification process of Fufang Tongmai macroporous resin were investigated by single factor test. Results: The optimal purification process was as follows: the mass concentration of the sample was 110 gL-1, the resin column height-height ratio was 1:10, the resin volume was 2.8:1 (adsorption time 3 h) BV 50% ethanol for elution. Conclusion: The purification process is reasonable and stable, which can be extended to large-scale production applications.
