目的:探讨杞菊胶囊对自发性高血压大鼠(SHR)左室肥厚及心肌原癌基因c-myc、N-ras表达的影响。方法:SHR随机分为4组:模型组、杞菊胶囊组、苯那普利组、杞菊胶囊加苯那普利组,以SD大鼠作为正常对照组。给药12周后取材,测量左室重量并以半定量逆转录-聚合酶链反应(RT-PCR)法测定各组大鼠心肌c-myc、N-ras的表达。结果:各组SHR心肌原癌基因c-myc、N-ras表达水平明显高于正常对照组(P<0.05),各治疗组较模型组均明显降低(P<0.05),杞菊胶囊与苯那普利疗效无统计学差异。结论:杞菊胶囊可防治心肌肥厚,其治疗机理可能与抑制心肌原癌基因c-myc、N-ras的高表达有关。
Objective: To investigate the effect of Qiju Capsule on the left ventricular hypertrophy and the expression of c-myc and N-ras genes in spontaneously hypertensive rats (SHR). Methods: SHR were randomly divided into 4 groups: model group, Qiju capsule group, benazepril group, Qiju capsule plus Benazepril group, and SD rats as normal control group. After 12 weeks of administration, the left ventricular mass was measured and semiquantitative reverse transcription-polymerase chain reaction (RT-PCR) was used to determine the expression of c-myc and N-ras in the myocardium of rats. Results: The expression levels of c-myc and N-ras in myocardium of SHR in each group were significantly higher than those in the normal control group (P<0.05). Compared with the model group, the expression of SHR in each group was significantly lower (P<0.05). Pregnancy efficacy was not statistically different. Conclusion: Qiju capsule can prevent and treat myocardial hypertrophy, and its therapeutic mechanism may be related to the inhibition of the high expression of c-myc and N-ras.
