果胶甲酯酶抑制因子(PMEI)能够抑制果胶甲酯酶(PME)的脱酯化作用,从而调控果胶甲酯酶的活性。本文在桃(Prunus persica)中鉴定出30个PpPMEI基因家族成员并进行了生物信息学分析,构建进化树发现,其与拟南芥PMEI基因家族进化关系较远。对PpPMEI进行蛋白性质分析发现,大多数PpPMEI蛋白存在信号肽,等电点在4.44~10.02之间,亚细胞定位表明多定位于分泌通路。部分PpPMEI表达模式分析表明,其在花、果实、叶中有表达差异,Prupe.1G118800和Prupe.5G112600在果实发育过程中表达量下降。赤霉素(GA)、脱落酸(ABA)和水杨酸(SA)三种激素处理后,Prupe.5G048900和Prupe.5G112600被GA和ABA诱导表达,Prupe.7G193700的表达被GA抑制。本文为研究PpPMEI在调控桃的花与果实发育以及激素响应等方面提供参考。
Pectin methylesterase inhibitory factor (PMEI) can inhibit pectin methylesterase (PME) deesterification, and thus regulate the activity of pectin esterase. In this paper, 30 members of PpPMEI gene family were identified in Prunus persica and analyzed by bioinformatics to construct phylogenetic tree, which has a far evolutionary relationship with Arabidopsis PMEI gene family. Protein characterization of PpPMEI revealed that most of the PpPMEI proteins had signal peptides with isoelectric points between 4.44 and 10.02, and subcellular localization indicated that they were localized in the secretory pathway. The analysis of partial PpPMEI expression patterns showed that the expression patterns of PpPMEI in flowers, fruits and leaves were different. The expression of Prupe.1G118800 and Prupe.5G112600 decreased during fruit development. After treatment with GA, ABA and SA, Prupe.5G048900 and Prupe.5G112600 were induced by GA and ABA, Prupe.7G193700 was inhibited by GA. This article provides a reference for the study of PpPMEI in the regulation of peach flower and fruit development and hormone response.
