目的:观察酪氨酸羟化酶(TH)、多巴胺脱羧酶(DDC)、多巴胺D2受体(DAD2R)在SD大鼠肠粘连模型中的表达,探讨多巴胺与术后肠粘连的关系。方法:选取SD雄性大鼠96只,随机分为3组:正常对照组(n=14),假手术组(n=42),肠粘连模型组(n=42),采用Ellis法制备肠粘连模型。假手术组及模型组分别在术后第2、5、8天分批处死,动物处死后取盲肠行免疫组化染色。结果:TH、DDC、DAD2R在模型组术后第2、5、8天的表达,较正常对照组及假手术组均有升高(P<0.05),随着时间的变化并逐渐降低。结论:术后肠粘连形成过程中,TH、DDC、DAD2R表达增高,提示多巴胺及其受体参与了术后早期肠粘连的形成。
Objective: To observe the expression of tyrosine hydroxylase (TH), dopamine decarboxylase (DDC) and dopamine D2 receptor (DAD2R) in the intestinal adhesion model of SD rats and explore the relationship between dopamine and postoperative intestinal adhesion. Methods: Ninety-six SD male rats were randomly divided into three groups: normal control group (n = 14), sham operation group (n = 42) and intestinal adhesion model group (n = 42) model. The sham operation group and model group were sacrificed on the 2nd, 5th and 8th day after operation. Immunohistochemical staining of the cecum was performed after sacrificed. Results: The expressions of TH, DDC and DAD2R in model group were significantly higher than those in normal control group and sham operation group on the 2nd, 5th and 8th day (P <0.05), and gradually decreased with time. Conclusion: The expression of TH, DDC and DAD2R is increased in the process of intestinal adhesion formation, suggesting that dopamine and its receptor are involved in the formation of early postoperative intestinal adhesion.
