目的调查贵州省不同地区虫媒病毒分布状况,对采集的蚊虫标本进行病毒分离与鉴定。方法 2008年7月在贵州省黔西、德江、榕江和从江县的4个标本采集点使用诱蚊灯采集蚊虫标本,通过组织培养法分离病毒,并对病毒分离物进行血清学和分子生物学鉴定;利用生物信息学技术对新分离病毒的序列进行分析,完成同源性和系统发生分析。结果采集到3属4种共计9160只蚊虫标本,从中分离到9株病毒,鉴定结果显示8株为盖塔病毒,1株为流行性乙型脑炎病毒(JEV);贵州省新分离盖塔病毒与原型株相比,核苷酸同源性为94.5%~94.9%,氨基酸同源性为97.4%~97.6%;新分离JEV与基因Ⅰ型JEV的同源性最高,系统进化分析结果显示属于基因Ⅰ型JEV。结论在贵州省分离到盖塔病毒和基因Ⅰ型JEV。盖塔病毒新分离株与我国其他省份分离株进化关系较近;JEV新分离株与四川省分离株进化关系较近。
Objective To investigate the distribution of arboviruses in different regions of Guizhou Province and to isolate and identify the collected mosquitoes. Methods In July 2008, mosquitoes were collected by mosquito-killing lamp in four specimens collection sites in Qianxi, Dejiang, Rongjiang and Congjiang counties of Guizhou province. The viruses were isolated by tissue culture and serological and molecular analysis of virus isolates Biological identification; the use of bioinformatics technology to analyze the sequence of the newly isolated virus to complete the homology and phylogenetic analysis. Results A total of 9160 mosquitoes were collected from 3 genera and 4 species, and 9 viruses were isolated from them. The results showed that 8 strains were Geeta virus and 1 strain was JEV. Compared with the original strain, the nucleotide homology was 94.5% ~ 94.9% and the amino acid homology was 97.4% ~ 97.6%. The highest homology was found between JEV and genotype Ⅰ JEV and phylogenetic analysis Belonging to genotype Ⅰ JEV. Conclusions Geeta virus and genotype Ⅰ JEV were isolated in Guizhou Province. The new GETTA isolates have close evolutionary relationship with other isolates in China. The new JEV isolates have close evolutionary relationship with Sichuan isolates.
