目的:研究细胞间粘附分子-1(ICAM-1)反义DNA对内皮细胞ICAM-1表达的抑制作用。方法:构建ICAM-1反义DNA载体,用脂质体转染内皮细胞,TNFα刺激后,流式细胞术检测转染细胞表面ICAM-1蛋白的表达。结果:酶切鉴定证明,1.4kb的ICAM-1DNA片段反向连接到pcDNA3表达载体;流式细胞术检测显示,正常细胞加TNFα刺激后,表面ICAM-1表达明显升高(P<0.01),转染细胞加TNFα刺激后表面ICAM-1表达无明显升高(P>0.05),转染细胞表面ICAM-1表达低于正常细胞(P<0.05)。结论:ICAM-1反义DNA体外可抑制细胞ICAM-1表达。
Objective: To investigate the inhibitory effect of ICAM-1 antisense DNA on ICAM-1 expression in endothelial cells. Methods: The ICAM-1 antisense DNA vector was constructed and transfected into endothelial cells by liposome. The expression of ICAM-1 on the surface of transfected cells was detected by flow cytometry. Results: The restriction endonuclease digestion proved that the 1.4kb ICAM-1 DNA fragment was reversely ligated into the pcDNA3 expression vector. The results of flow cytometry showed that ICAM-1 expression was significantly increased in normal cells stimulated with TNFα (P <0. 01). The expression of ICAM-1 on the transfected cells plus TNFα was not significantly increased (P> 0.05). The expression of ICAM-1 on the transfected cells was lower than that on the normal cells (P <0.05). Conclusion: ICAM-1 antisense DNA can inhibit ICAM-1 expression in vitro.
