摘 要:对建立猪和牛的胚胎干细胞系(Embryonic stem cells,ESCs)的条件进行了摸索和优化,建立了良好的胚胎培养体系、内细胞团分离方法和饲养层的选择;另外分别建立猪和牛的诱导性多能性干细胞系(induced pluripotent stem cells,iPS),并分别对其进行了鉴定;对目前的培养体系进行了优化提高了iPS的效率和发育潜能;尝试以iPS为供体细胞进行核移植,以期获得iPS来源的动物种群。

Abstract:Exploration and optimization the conditions for establishment of pigs and cattle embryonic stem cell lines (ESCs) to establish a good embryo culture system, the inner cell mass separation methods and choice of feeder layers. Established pigs and cattle induced pluripotent stem cells (iPSCs), with respect to their identification; the current training system has been optimized to improve the efficiency and growth potential of iPSCs; try to obtain iPS-derived animal populations, by nuclear transfer that iPS cells as donor cells.
