目的:探讨黄芩苷对宫颈癌细胞放射敏感性的影响及作用机制。方法:体外培养人宫颈癌Hela细胞,分别转染pcDNA(+)空载体和pcDNA-UCA1,对正常培养细胞和转染后细胞进行黄芩苷处理或黄芩苷联合放射处理,MTT实验检测细胞增殖抑制率,克隆形成实验检测放疗敏感性,流式细胞术检测细胞凋亡情况。结果:黄芩苷呈剂量-时间依赖性抑制Hela细胞增殖;经黄芩苷处理,Hela细胞在不同剂量照射下的细胞存活分数呈照射剂量依赖性下降;黄芩苷和放射处理均能促进Hela细胞凋亡,其中黄芩苷联合放射对Hela凋亡的促进作用最明显;黄芩苷能够抑制放射下Hela细胞中lncRNA UCA1的表达,而过表达lncRNA UCA1则逆转了黄芩苷对Hela细胞放射敏感性和细胞凋亡的促进作用。结论:黄芩苷可通过抑制lncRNA UCA1的表达,促进宫颈癌细胞Hela的放射敏感性。“,”Objective:To investigate the effect of baicalin on radiosensitivity of cervical cancer cells and its mechanism.Methods:Human cervical cancer Hela cells were cultured in vitro and transfected with pcDNA (+) empty vector or pcDNA-UCA1. Normal-cultured cells and transfected cells were then treated with baicalin with or without radiation treatment. MTT assay was used to detect the inhibition rate of cell proliferation, clone formation assay was used to measure the radiosensitivity, and flow cytometry was used to examine cell apoptosis.Results:Baicalin inhibited the proliferation of Hela cells in a dose-time dependent manner. After baicalin treatment, the surviving fraction of Hela cells under different radiation doses decreased in a dose-dependent manner. Both baicalin and radiation treatment promoted Hela cell apoptosis, and such effect on Hela cell apoptosis was most prominent with combined use of baicalin and radiation treatment. Baicalin could inhibit the expression of lncRNA UCA1 in Hela cells under radiation. Overexpression of lncRNA UCA1 reversed the effect of baicalin on increasing radiosensitivity and apoptosis of Hela cells.Conclusion:Baicalin can promote the radiosensitivity of cervical cancer Hela cells by inhibiting the expression of lncRNA UCA1.
