以紫花苜蓿、木豆、大豆叶片及大豆种子为试验材料,采用Trizon法、改良CTAB法及试剂盒法提取样品总RNA,并比较3种提取方法的提取效果.试验结果表明:试剂盒法提取的4种材料总RNA的28S、18S条带清晰完整,A260/A280比值在1.9~2.0之间,而其他两种方法所获得的总RNA产量较低,且纯度不高.由此可见,试剂盒法能够高效地提取紫花苜蓿、木豆、大豆叶片及大豆种子的总RNA,为以后的分子水平研究打下基础.“,”This experiment aimed at comparing the extracting result amount Trizon method,improved CTAB method and kit method which about the total RNA extracting from alfalfa,pigeonpea,soybean leaves,soybean seeds.The extracting results showed that kit method extracted the total RNA of 28S,18S clear and complete bands by 4 materials.Ratio of A260/A280 was between 1.9~2.0,but the total RNA extracted from other two methods had lower yield and purity.Therefore,kit method could efficiently extract total RNA from alfalfa,pigeonpea,soybean leaves,soybean seeds,which laid the foundation of molecular research.
