目的:端粒酶活性存在于人类几乎所有永生细胞株中,放射治疗仍是目前口腔癌综合治疗的重要组成部分,本研究旨在探讨X线对人舌鳞状细胞癌系Tca8113细胞端粒酶活性的影响。方法:体外培养的人舌鳞癌细胞系Tca8113经0、2、4、8、12、20 Gy剂量X线照射,于照射后0.5 h、1 h、3 h、5 h收集细胞,采用TRAP法检测不同剂量X线照射前后Tca8113细胞中端粒酶活性。结果:未经X线照射时,Tca8113细胞呈端粒酶阳性,照射后仅2 Gy组存在端粒酶活性,其它各照射剂量组细胞均呈端粒酶阴性。结论:人舌鳞癌细胞系Tca8113是端粒酶阳性细胞系,端粒酶活性是Tca8113细胞经放射线照射后赖以生存的重要因素;Tca8113细胞的端粒酶活性具有放射剂量依赖性,经X线照射后,端粒酶活性显著下调;2 Gy是体外Tca8113细胞能够耐受的单次照射剂量。
Objective: Telomerase activity exists in almost all human immortalized cell lines. Radiation therapy is still an important part of comprehensive oral cancer treatment. The purpose of this study is to investigate the effect of X-ray on telomerase activity in human tongue squamous cell carcinoma Tca8113 cells Effect of activity. Methods: Human tongue squamous cell carcinoma cell line Tca8113 was irradiated with 0, 2, 4, 8, 12 and 20 Gy dose X-rays. Cells were harvested at 0.5 h, 1 h, 3 h and 5 h after irradiation. The telomerase activity in Tca8113 cells before and after different doses of X-ray irradiation was detected. Results: Tca8113 cells were positive for telomerase without X-ray irradiation. Telomerase activity was only found in 2 Gy group after irradiation, and telomerase negative cells were found in other irradiation dose groups. Conclusion: Human tongue squamous cell carcinoma cell line Tca8113 is a telomerase-positive cell line, telomerase activity is an important factor for Tca8113 cells to survive after being irradiated by radiation. The telomerase activity of Tca8113 cells is dose- Telomerase activity was significantly down-regulated after irradiation; 2 Gy was a single radiation dose that Tca8113 cells could tolerate in vitro.
