对输精管平滑肌中分离的蛋白激酶C(PKC)抑制剂的生理作用进行了研究。应用放射自显形方法证明、此抑制剂对输精管平滑肌中PKC底物-40KD蛋白的磷酸化有明显抑制作用;并用免疫印迹电泳法确定了输精管中PKC亚型为ε亚型。以上结果在明,输精管平滑肌中PKC抑制剂通过与PKC-ε的相互作用,参与对平滑肌舒张与收缩的调控,还可能参与细胞分泌等细胞运动功能。
Physiological effects of protein kinase C (PKC) inhibitors isolated from vas deferens smooth muscle were studied. Autoradiography demonstrated that this inhibitor significantly inhibited the phosphorylation of -40KD protein, a PKC substrate in vas deferens smooth muscle. PKC subtypes were identified as ε subtype in vas deferens by immunoblotting. The above results show that PKC inhibitors in the vas deferens smooth muscle participate in the regulation of diastole and contraction of smooth muscle by interacting with PKC-ε, and may also participate in cell motility such as cell secretion.
