以感染郁金香碎锦病毒(TBV)和百合无症病毒(LSV)的O/T系百合黄天霸(Manissa)为试验材料,进行了茎尖诱导培养基的筛选;采用不同大小茎尖培养和热处理结合茎尖培养的方式脱除病毒;运用RT-PCR技术进行病毒检测。结果表明,MS+6-BA0.5mg/L+NAA0.3mg/L培养基比较适宜茎尖不定芽的诱导;(39±1)℃热处理结合0.5~0.7 mm大小的茎尖培养脱毒效果最好。
The tip induction medium was screened from O / T-lily Mantibae infected with tulipavirus (TBV) and lily disease-free virus (LSV) The virus was removed by heat treatment in combination with the shoot tip culture. The virus was detected by RT-PCR. The results showed that MS + 6-BA0.5mg / L + NAA0.3mg / L medium was suitable for induction of adventitious shoots at the shoot tip. The heat treatment of (39 ± 1) ℃ combined with 0.5 ~ 0.7mm shoot tips it is good.
