目的:建立明党参愈伤组织的诱导及其细胞悬浮培养的技术体系。方法:采用正交试验法研究不同植物激素对明党参叶柄、根和叶片愈伤组织诱导的影响,以及明党参不同外植体细胞的悬浮培养。结果:叶柄最易诱导出愈伤组织,其次是根和叶片,4种植物激素对明党参根和叶柄愈伤组织的诱导都具有极显著的效果,其中诱导叶柄愈伤组织形成的最佳激素组合为0.5 mg.L-1NAA+0.5 mg.L-12,4-D+0.1 mg.L-1KT+0.5 mg.L-16-BA,培养14 d左右,诱导率达到100%。此外,明党参叶柄细胞培养至21 d时,细胞量和多糖均达到最大值,分别1 090.28,36.85 mg.L-1。结论:明党参叶柄是愈伤组织诱导的最佳外植体和细胞悬浮培养体系的最佳细胞来源。
Objective: To establish the technology system of callus induction and cell suspension culture of Codonopsis Pilosula. Methods: The effects of different plant hormones on callus induction of petiole, root and leaf of Ming dang ginseng were studied by orthogonal test and the suspension culture of cells of different explants of Codonopsis pilosula. Results: The petioles were most likely to induce callus, followed by the roots and leaves. The four plant hormones had a very significant effect on the induction of callus and the callus of the petiole. The best hormones that induced the formation of petiole callus The combination was 0.5 mg.L-1NAA + 0.5 mg.L-12,4-D + 0.1 mg.L-1KT + 0.5 mg.L-16-BA, and the induction rate reached 100% after about 14 days. In addition, the cell number and polysaccharides all reached the maximum value when they were cultured for 21 days, which were respectively 1 090.28,36.85 mg.L-1. Conclusion: Petiole of Codonopsis pilosula is the best cell source for callus induction of explants and cell suspension culture system.
