目的探讨白藜芦醇对糖尿病大鼠肾功能保护作用的机制。方法一次性注射链脲佐菌素法建立大鼠糖尿病肾病(DN)模型,生化分析法检测血糖,酶联免疫法测定24 h尿白蛋白排泄率,肌酐清除率,实时多聚酶链反应(RT-PCR)法检测线粒体分裂、融合基因转录水平变化,检测线粒体膜电势。结果白藜芦醇治疗对DN大鼠血糖无明显影响(P>0.05),但可明显降低肌酐清除率和24 h尿白蛋白量(P<0.05),降低线粒体分裂融合相关基因Mfn1、Fis1、Drp1 mRNA表达,提高线粒体膜电势(P<0.05)。结论白藜芦醇可通过影响线粒体动态平衡和功能发挥对DN大鼠肾脏功能的保护作用。
Objective To investigate the protective effect of resveratrol on renal function in diabetic rats. Methods A rat model of diabetic nephropathy (DN) was established by a single injection of streptozotocin (STZ) and the blood glucose was measured by biochemical analysis. The urinary albumin excretion rate, creatinine clearance rate and real-time polymerase chain reaction (RT- PCR) was used to detect the changes of mitochondrial fission and fusion gene transcription, and the mitochondrial membrane potential was detected. Results Resveratrol had no significant effect on blood glucose in DN rats (P> 0.05), but could significantly reduce creatinine clearance and 24 h urinary albumin (P <0.05), decrease mitochondrial fission fusion related genes Mfn1, Fis1, Drp1 mRNA expression increased mitochondrial membrane potential (P <0.05). Conclusion Resveratrol can protect renal function of DN rats by affecting the dynamic balance and function of mitochondria.
