目的 应用新型核酸荧光染料Sybr-GreenI检测端粒酶活性,并探讨该酶活性在结直肠癌临床诊断中的意义。方法 用端粒重复扩增 (TRAP)结合Sybr GreenI技术检测 4 6例结直肠癌及 1 9例癌旁组织中端粒酶的活性。结果 Sybr-GreenI能清晰显示端粒酶扩增后的条带。端粒酶活性在结直肠癌中阳性率为 8 9. 1 3% ,在癌旁组织中未发现端粒酶活性。端粒酶活性与细胞分化程度、Dukes分期、有无淋巴结转移均无关。结论 用Sybr-GreenI检测端粒酶活性对于诊断结直肠癌具有重要的临床辅助价值。
Objective To detect the telomerase activity by a new fluorescent dye Sybr-GreenI and to explore its significance in the clinical diagnosis of colorectal cancer. Methods telomeric repeat amplification (TRAP) combined with Sybr GreenI was used to detect telomerase activity in 46 cases of colorectal cancer and 19 cases of paracancerous tissues. Results Sybr-GreenI clearly shows telomerase amplified bands. The positive rate of telomerase activity in colorectal cancer was 8 9.13%. No telomerase activity was found in adjacent tissues. Telomerase activity and cell differentiation, Dukes staging, with or without lymph node metastasis. Conclusion Sybr-GreenI detection of telomerase activity for the diagnosis of colorectal cancer has important clinical value.
