目的 探讨咳喘落协同地塞米松拮抗支气管哮喘气道变应性炎症作用的分子机制。方法 采用逆转录 聚合酶链反应 (RT PCR)半定量地测定了不同剂量的咳喘落对哮喘大鼠肺组织和外周血单个核细胞(PBMC)中IL 4mRNA、IL 5mRNA表达的影响。结果 哮喘时大鼠肺组织和PBMC中IL 4mRNA与IL 5mRNA表达均增强 ,显然高于空白对照组 (P <0 .0 1) ,经大剂量咳喘落治疗后 ,其表达同时显著降低 (P <0 .0 5 ,P <0 .0 1) ,与地塞米松组无差异 (P >0 .0 5 ) ,小剂量咳喘落治疗对哮喘大鼠肺组织中IL 4mRNA的表达有显著的抑制作用 (P <0 .0 5 ) ,但小剂量咳喘落加地塞米松对哮喘大鼠肺组织和PBMC中IL 4mRNA、IL 5mRNA表达的抑制显著强于大剂量咳喘落和地塞米松 (P <0 .0 5 ,P <0 .0 1)。结论 咳喘落能够同时降低大鼠肺组织和PBMC中IL 4和IL 5基因转录 ,减少其生物合成 ,进而抑制嗜酸性粒细胞的聚集、活化 ,发挥其抗气道炎症的作用 ,与地塞米松合用有协同作用
Objective To investigate the molecular mechanism of Kechuanluo combined with dexamethasone against airway allergic inflammation in bronchial asthma. Methods RT-PCR was used to semi-quantitatively determine the effects of different doses of Kechuanluo on the expression of IL-4mRNA and IL-5mRNA in lung and PBMC of asthmatic rats. Results The expressions of IL-4 mRNA and IL-5 mRNA in lung tissue and PBMC of asthmatic rats were significantly increased after asthma, which were significantly higher than those of the blank control group (P <0.01). The expression of IL-4 mRNA and IL- <0.05, P <0.01), there was no difference between dexamethasone group and dexamethasone group (P> 0.05), the expression of IL-4mRNA in lung tissue of asthmatic rats was significantly decreased (P <0.05). However, the inhibition of IL-4mRNA and IL-5 mRNA expression in asthmatic rat lungs and PBMCs with low-dose Kechuanluo plus dexamethasone was significantly stronger than that of high-dose Kechuanluo and dexamethasone P <0. 05, P <0. 01). Conclusions Kechuan Liao can reduce the transcription of IL 4 and IL 5 genes in lung and PBMCs of rats simultaneously, decrease their biosynthesis, inhibit the aggregation and activation of eosinophils and exert their anti-airway inflammation, Sycamore synergy
