目的研究复方曲肽对大鼠缺血再灌注损伤的保护作用。方法将雄性SD大鼠随机分为正常对照(normal control,NC)组、大脑中动脉闭塞再灌注(middle cerebral artery occlusion reperfusion,MCAO/R)组及MCAO/R+复方曲肽[MCAO/R+(troxerutin and cerebroprotein,TC)]组3组(每组10只)。缺血1 h再灌注23 h后,各组取6只动物接受神经功能评分后进行MRI检查,随后处死行2,3,5-三苯基氯化四氮唑染色检测脑梗死体积;各组另4只动物处死,免疫蛋白印迹检测缺血半暗带中含半胱氨酸的天冬氨酸蛋白水解酶1(cysteinyl aspartate specific proteinase 1,Caspase 1)、Caspase 3及Caspase 8的表达。结果 MCAO/R组术后动物均出现了脑梗死病灶及神经功能的损伤,复方曲肽治疗可减小脑梗死体积并改善神经功能。MCAO/R组缺血半暗带中Caspase 1、Caspase 3、Caspase 8表达显著高于NC组(P<0.05),MCAO/R+TC组Caspase 1、Caspase 3、Caspase 8表达低于MCAO/R组(P<0.05)。结论复方曲肽对缺血再灌注损伤具有明显的保护作用,其机制与复方曲肽抑制细胞凋亡相关基因表达从而达到抗凋亡作用有关。
Objective To study the protective effect of compound curcumin on ischemia-reperfusion injury in rats. Methods Male SD rats were randomly divided into normal control (NC) group, middle cerebral artery occlusion reperfusion (MCAO / R) group and MCAO / R + triptorelin (MCAO / R + and cerebroprotein (TC)] group (group 10). After ischemia for 1 h and reperfusion for 23 h, 6 animals in each group received neurological function score and MRI examination, and then were sacrificed 2,3,5-triphenyltetrazolium chloride tetrazolium staining for detection of cerebral infarction volume; each group Four other animals were sacrificed and the expression of cysteinyl aspartate specific proteinase 1 (Caspase 1), Caspase 3 and Caspase 8 in ischemic penumbra was detected by Western blotting. Results All the animals in the MCAO / R group had lesions of cerebral infarction and neurological damage. The treatment with the complex curcumin decreased the volume of cerebral infarction and improved the neurological function. The expression of Caspase 1, Caspase 3 and Caspase 8 in ischemic penumbra of MCAO / R group was significantly higher than that of NC group (P <0.05), while the expression of Caspase 1, Caspase 3 and Caspase 8 in MCAO / R + TC group was lower than that of MCAO / R Group (P <0.05). Conclusion Compound curcumol has a significant protective effect on ischemia-reperfusion injury. Its mechanism is related to the inhibition of apoptosis-related gene expression by the compound melittin so as to achieve the anti-apoptotic effect.
