目的:提出了采用固相萃取-高效液相色谱法同时测定孕妇血清中四种环境内分泌干扰物的含量。方法:以OASIS HLB小柱提取血清中的BPA,NP,p,p’-DDE和DEHP,采用Kromasil C18色谱柱(4.6 mm×250 mm,5μm)分离,流动相为甲醇和水(95∶5,v/v),采用荧光检测器和二极管阵列检测器同时检测。结果:BPA和NP在1 ng/ml~250 ng/ml范围内线性关系良好,p,p’-DDE和DEHP在10 ng/ml~1000 ng/ml范围内线性关系良好;方法回收率在94.8%~101.6%之间,相对标准偏差在3.8%~9.0%之间。结论:该方法样品处理简单、操作简便,能够节省生物样品,缩短检测时间,提高检测效率,可用于血清中BPA、NP、p,p’-DDE和DEHP的定性、定量分析。
OBJECTIVE: To simultaneously determine the content of four environmental endocrine disruptors in pregnant women’s serum by solid phase extraction-high performance liquid chromatography. Methods: The serum samples of BPA, NP, p, p’-DDE and DEHP were extracted with OASIS HLB cartridge and separated on a Kromasil C18 column (4.6 mm × 250 mm, 5 μm). The mobile phase consisted of methanol and water , v / v) using a fluorescence detector and a diode array detector. Results: There was a good linear relationship between BPA and NP in the range of 1 ng / ml to 250 ng / ml. The linearity of p, p’-DDE and DEHP in the range of 10 ng / ml to 1000 ng / ml was good. % ~ 101.6%, relative standard deviation between 3.8% ~ 9.0%. Conclusion: The method has the advantages of simple processing and simple operation, which can save biological samples, shorten the detection time and improve the detection efficiency, and can be used for qualitative and quantitative analysis of BPA, NP, p, p’-DDE and DEHP in serum.
