目的 :探讨中性粒细胞 (PMN)与血管内皮细胞 (VEC)的粘附特性和影响因素 ,以及中药复圣散的干预作用。方法 :以培养人脐静脉内皮细胞作为靶细胞 ,观察在高脂、高糖、肿瘤坏死因子 (TNF -α)及缺氧再氧化条件下 ,对培养的VEC活性以及PMN与VEC相互粘附的影响。结果 :高脂、高糖、TNF -α和缺氧再氧化等均可抑制VEC活性 ,并显著增强PMN与VEC的相互粘附作用 ,其粘附率正常对照组为 1 0 0 % ,高脂组则为 1 67 7% ,缺氧再氧化组为1 5 1 4% ,TNF -α组为 1 3 6 7% ,高糖组为 1 2 9 6% (均P <0 0 1 )。如事先在试验体系中加入不同浓度的中药血清 ,则粘附率明显降低 ,大、小剂量均呈现有抑制粘附效应 ,并有统计学差异。结论 :中药复圣散有良好的保护VEC和抗粘附作用 ,这可能是该方防治缺血性心脑血管疾病的机制之一。
Objective: To investigate the adhesion characteristics and influencing factors of neutrophils (PMN) and vascular endothelial cells (VEC), and the intervention of Fusheng San. METHODS: Cultured human umbilical vein endothelial cells were used as target cells to observe the effects of high-fat, high-sugar, tumor necrosis factor (TNF-α), and hypoxia-reoxidation conditions on the activity of cultured VECs and the mutual adhesion between PMNs and VECs. influences. Results: High fat, high glucose, TNF-α and hypoxia reoxidation all inhibited the activity of VEC and significantly enhanced the mutual adhesion between PMN and VEC. The normal rate of adhesion was 100% in the control group. The group was 167.7%, the anoxia-reoxygenation group was 151.4%, the TNF-α group was 136.7%, and the high glucose group was 129.6% (all P0.01). If different concentrations of Chinese herbal serum were added to the test system in advance, the adherence rate was significantly reduced, and both large and small doses exhibited inhibitory effects on adhesion, with statistical differences. Conclusion : Chinese medicine Fusheng San has good protection of VEC and anti-adhesion effect, which may be one of the mechanisms of the prevention and treatment of ischemic cardiovascular and cerebrovascular diseases.
