目的:观察糖原合成酶激酶-3β(glycogen synthase kinase,GSK3β)在大鼠慢性移植物肾病(chronic allograft nephropathy,CAN)模型移植肾中的表达。探讨GSK3β在CAN中的作用。方法:F344大鼠为供体,Lewis大鼠为受体,行原位异体肾移植,建立大鼠CAN模型,术后12周处死各组动物,取移植肾标本观察组织形态学变化,并用SP免疫组织化学方法对移植肾组织GSK3β、NF-κBp65的表达进行观察。结果:与同系移植组、对照组相比,CAN组GSK3β、NF-κBp65在移植肾中的表达明显增强。结论:提示GSK3β可能在CAN的发病机制中发挥重要作用。
Objective: To observe the expression of glycogen synthase kinase (GSK3β) in renal allograft nephropathy (CAN) model rats. To investigate the role of GSK3β in CAN. METHODS: F344 rats were used as donors. Lewis rats were used as recipients. Allogeneic kidney transplantation was performed to establish a rat model of CAN. All animals were sacrificed at 12 weeks after operation. The morphological changes of the rats were observed by SP method. The expression of GSK3β and NF-κBp65 in renal allograft tissues was observed by immunohistochemistry. Results: Compared with the homologous transplantation group and the control group, the expression of GSK3β and NF-κBp65 in CAN group was significantly increased. Conclusion: It suggests that GSK3β may play an important role in the pathogenesis of CAN.
