目的:从脑细胞周期的角度揭示熟地黄防治脑衰老的作用机制。方法:36只SD大鼠随机分为正常组、模型组、天保宁组和熟地黄组,采用D-半乳糖造模同时服用相应药物45d。在检测大鼠行为学、脑组织生化指标、细胞衰老特异代谢产物SA-β-gal阳性表达细胞数的基础上,采用流式细胞术检测细胞周期、免疫组化法检测海马CA1区P53,P21,P16的表达。结果:熟地黄能一定程度促进细胞周期从G0/G1期向S期进展,抑制海马CA1区CDK抑制基因蛋白P53,P21,P16的表达。结论:熟地黄能调控细胞周期相关基因P53,P21,P16表达。
OBJECTIVE: To reveal the protective mechanism of Rehmannia glutinosa on brain aging from the perspective of brain cell cycle. Methods: Thirty-six SD rats were randomly divided into normal group, model group, Tianbaoning group, and Rehmannia group. The rats were treated with D-galactose for 45 days. The cell cycle was detected by flow cytometry and the P53 and P21 in the hippocampal CA1 area were detected by immunohistochemistry on the basis of ethology, brain tissue biochemical markers and SA-β-gal positive expression cell number. P16 expression. Results: Rehmannia glutinosa could promote the progression of cell cycle from G0/G1 phase to S phase, and inhibit the expression of CDK inhibitory gene proteins P53, P21 and P16 in hippocampal CA1 region. Conclusion: Rehmannia glutinosa can regulate the expression of cell cycle related genes P53, P21 and P16.
