Objective To investigate the levels of endothelin (ET) in plasma and liver tissue during obstructive jaundice (OJ) formation and its role in OJ hepatic injury. Methods The levels of ET in plasma and liver tissue were determined at 3, 6, 9, 1, 2 and 5 days after BDL by radioimmunas (RIA) in rat model of Bile Ductligation (BDL) At the same time, the change of serum ALT level was observed and compared with the pathological changes of liver tissue. Results Serum total bilirubin (TBi) increased rapidly after BDL, which was significantly higher than that of normal group and sham operation group (P <0.01). ET level of plasma and liver tissue of BDL group was significantly higher than that of control group <0 0 1). There was significant difference between BDL groups (F = 1 8 4 1, P 0 01 and F 0 4 01 1 1, P 0 01), with the prolongation of obstruction time The content of serum ETT was the highest at 15d, which were (2 94 98 ± 3595) ng / L and (2018 ± 2445) ng / 100g, respectively. (r = 0 84 41, P <0 01 and r 0 93 0 3, P 0 01). Compared with BDL group, the content of ET in BDL + ET -Ab group was lower than that in BDL group (P <0.05) The level also decreased (P <0.01). The results suggest that the content of ET in the blood and liver tissue increased in OJ and increased with the prolongation of obstruction time. ET -Ab could reduce the content of ET and reduce the liver injury. ET may be one of the important factors involved in OJ liver injury. A large number of small bile duct hyperplasia may be associated with OJ E
