1 材料和方法 1.1 材料 Tyrphostin-51、PD098059、N-羟乙基哌嗪-N’-2-乙磺酸(HEPES)、钒酸钠(Na_3VO_4)、亮抑蛋白酶肽(leupeptin)、抑蛋白酶肽(aprotinin)、1,10-菲咯啉(1,10-phenanthroline)、二硫苏糖醇(DTT)、精胺、亚精胺、NP-40、三硝基甲苯(Triton X-100)和苯甲基磺酰氟(PMSF)均购自美国Sigama公司,蛋白质定量检测试剂盒(Protein assay kit)购自美国Bio-Rad公司,α-~(32)p-UTP和γ-~(32)p-ATP购自北京亚辉生物试剂公司,2,5-二苯基唑(PPO)和1,4-双-[5-酚-2-恶唑]-苯(POPOP)购自上海化学试剂厂,其他试剂为国产分析纯。LS6500型闪烁仪为美国Beckman公司产品,透析袋购自北京东方试剂仪器公司,(?)成年SD大鼠(220g～250g,第一军医大学动物所提供),禁食24h,以4mL(20mmol/L)去氧胆酸钠灌胃损伤胃粘膜,应用PTK抑制剂Tyrphostin-51(300μg·kg~(-1))和MAPKK抑制剂
Materials and Methods 1.1 Materials Tyrphostin-51, PD098059, N-hydroxyethylpiperazine-N’-2-ethanesulfonic acid (HEPES), sodium vanadate (Na3VO4), leupeptin, aprotinin aprotinin, 1,10-phenanthroline, dithiothreitol (DTT), spermine, spermidine, NP-40, Triton X-100 and (PMSF) were purchased from Sigama, USA. The protein assay kit was purchased from Bio-Rad, Inc., USA, α- (32) p-UTP and γ- p-ATP was purchased from Beijing Yahui Bio-Reagent Company, and 2,5-diphenyl azole (PPO) and 1,4-bis- [5-phenols-2-oxazole] Factory, other reagents for domestic analytical grade. The LS6500 scintillometer was purchased from Beckman Company in USA. The dialysis bag was purchased from Beijing Orient Reagent Instrument Co., Ltd. (220g ~ 250g, provided by the First Military Medical University) for 24 h.
