目的探讨建立最佳2型糖尿病合并脂肪肝大鼠模型方案。方法用不同方案给予链脲佐菌素、烟酰胺及高脂饲料干预大鼠8周后,制备脂肪肝动物模型。尾静脉取血,测定空腹血糖(FPG),全自动生化分析仪检测总胆固醇(TC)、三酰甘油(TG)、肝和血清谷丙转氨酶(ALT)、谷草转氨酶(AST);放免法检测空腹胰岛素(FINS),计算肝指数及胰岛素抵抗指数;光镜下观察肝组织改变。结果先注射链脲佐菌素50 mg·kg-1、高脂饲料喂养后,大鼠血糖、血脂、肝功能有显著的变化,组织病理学观察肝细胞出现脂变。结论先注射STZ后高脂饲料喂养可以成功复制糖尿病并脂肪肝动物模型,且符合人类此病的发病特点。
Objective To investigate the optimal rat model of type 2 diabetes mellitus complicated with fatty liver. Methods Different regimens were given streptozotocin, nicotinamide and high-fat diet for 8 weeks after the preparation of fatty liver model. Blood samples were taken from the tail vein and fasting blood glucose (FPG) was measured. Total cholesterol (TC), triglyceride (TG), liver and serum ALT and AST were detected by automatic biochemical analyzer. Fasting insulin (FINS), liver index and insulin resistance index were calculated. Liver tissue was observed under light microscope. Results The first injection of streptozotocin 50 mg · kg-1, high-fat diet, the rats blood glucose, blood lipids, liver function had significant changes in histopathology observed hepatocellular lipidosis. CONCLUSIONS: After the first injection of STZ, high-fat diet can successfully replicate the model of diabetic and fatty liver and is consistent with the onset of human disease.
