目的观察香连金黄散外用对皮肤炎症微环境的影响。方法采用免疫组化方法测定香连金黄散对阳证疮疡模型动物感染炎症皮肤IL-1、IL-6、IL-8、TNF-α、IFN-γ蛋白含量的影响。结果与模型对照组比较,香连金黄散高剂量组豚鼠皮肤组织中的白介素1蛋白表达明显降低(P<0.01);香连金黄散中剂量组豚鼠皮肤组织中的白介素6蛋白表达明显降低(P<0.01);香连金黄散高、中剂量组豚鼠皮肤组织中的白介素8蛋白表达降低(P<0.05);香连金黄散高、中剂量组豚鼠皮肤组织中的肿瘤坏死因子蛋白表达降低(P<0.05);香连金黄散高、中剂量组豚鼠皮肤组织中的干扰素-γ蛋白表达有所降低,但差异无统计学意义(P>0.05)。结论香连金黄散可能是通过下调感染皮肤局部的炎症因子表达而发挥其清热解毒、消肿止痛作用的。
Objective To observe the effect of Fragrant Golden Powder on the microenvironment of skin inflammation. Methods Immunohistochemical method was used to determine the effect of Xianglian Jinshuang San on the protein levels of IL-1, IL-6, IL-8, TNF-α and IFN-γ in inflammatory skin infected by Yang syndrome in rats. Results Compared with the model control group, interleukin-1 protein expression in guinea pig skin was significantly decreased (P <0.01), and the interleukin-6 protein expression in guinea pig skin was significantly decreased P <0.01). The expression of IL-8 in skin of Guinea pigs with high, middle and high dose of Xianglianjinhuang powder was decreased (P <0.05), while the expression of tumor necrosis factor (P <0.05). The expression of IFN-γ in the skin of Guinea pigs with high, medium and high doses of Fragrant Golden Powder had no significant difference (P> 0.05). Conclusions Honglian Jinshuang San may exert its detoxification, swelling and analgesic effects by down-regulating the expression of inflammatory cytokines in the skin.
