目的:观察养阴益气活血方药对干燥综合征NOD小鼠颌下腺组织Fas/FasL及其mRNA表达的影响。方法:32只NOD小鼠随机分为模型组、中药组(每天按100 g·kg-1灌胃养阴益气活血煎剂0.4 mL)、羟氯喹组(每天按60 mg·kg-1灌胃羟氯喹0.4 mL)、中西药组(每天灌胃养阴益气活血煎剂50 g·kg-1+羟氯喹60 mg·kg-1,0.4 mL),每组8只,选8只Balb/C小鼠做为正常对照组。实验进行8周后处死小鼠,解剖摘取颌下腺,免疫组织化学法检测颌下腺组织Fas,FasL蛋白的表达;RT-PCR法检测颌下腺组织Fas,FasL mRNA的表达。结果:模型组小鼠颌下腺组织Fas,FasL表达均高于其他各组(P<0.05)。对照组FasL表达明显低于羟氯喹组(P<0.05)。模型组小鼠颌下腺Fas mRNA相对表达量均高于其他组,对照组明显低于其他各组(P<0.05)。模型组小鼠颌下腺FasL mRNA表达高于中药组、中西药组(P<0.05);中西药组表达明显低于羟氯喹组(P<0.05)。结论:养阴益气活血煎剂能下调干燥综合征NOD小鼠颌下腺Fas,FasL及其mRNA的表达,且与羟氯喹合用效果较好。养阴益气活血煎剂可能通过减少Fas/FasL调控的细胞凋亡达到治疗SS的作用。
Objective: To observe the effect of Yangyin Yiqi Huoxue Recipe on Fas / FasL and its mRNA expression in submandibular gland of NOD mice with Sjogren ’syndrome. Methods: Thirty-two NOD mice were randomly divided into model group, Chinese medicine group (0.4 mL daily by 100 g · kg-1 gavage yin yin qi and activating blood decoction), hydroxychloroquine group (60 mg · kg -1 irrigation daily (N = 30), and the control group (n = 10). The rats in the Chinese and western medicine groups were fed with Yin Yin Yiqi Huoxue Decoction 50 g · kg-1 and hydroxychloroquine 60 mg · kg-1, / C mice as normal control group. Mice were sacrificed 8 weeks after the experiment, the submandibular glands were dissected and the expressions of Fas and FasL protein in submandibular gland were detected by immunohistochemical method. The expressions of Fas and FasL mRNA in submandibular gland were detected by RT-PCR. Results: The expressions of Fas and FasL in model group were higher than those in other groups (P <0.05). The expression of FasL in control group was significantly lower than that in hydroxychloroquine group (P <0.05). The relative expression of Fas mRNA in the submandibular gland of the model group was higher than that of the other groups, and the control group was significantly lower than the other groups (P <0.05). The expression of FasL mRNA in submandibular gland of model group was higher than that of traditional Chinese medicine group and western medicine group (P <0.05). The expression of FasL mRNA in submandibular gland of model group was lower than that of hydroxychloroquine group (P <0.05). Conclusion: Yangyin Yiqihuoxue Decoction can reduce the expression of Fas, FasL and its mRNA in the submandibular gland of the mice with Sjogren ’s syndrome, and it has good effect with hydroxychloroquine. Yangyin Yiqihuoxue decoction may achieve the effect of treating SS by reducing the Fas / FasL-regulated apoptosis.
