目的:研究江苏地产白首乌总苷对四氯化碳所致大鼠肝纤维化的保护作用。方法:Wistar大鼠随机分6组,正常组、模型组给1%CMC-Na,阳性药组给氢化可的松3.5 mg.kg-1,白首乌低、中、高剂量组分别给予白首乌总苷4,8,16 mg.kg-1。大鼠每周2次灌胃50%CCl4花生油2 mL.kg-1,连续8周。造模与给药同时进行。8周后取血,计算大鼠的肝脾指数,检测大鼠血清中谷丙转氨酶(GPT),谷草转氨酶(GOT)的活力;检测血清中透明质酸(HA)、Ⅲ型前胶原(PCⅢ)的含量;检测肝脏组织中超氧化物歧化酶(SOD)活性、羟脯氨酸(HyP)和丙二醛(MDA)含量。观察大鼠肝组织病理变化。结果:江苏地产白首乌总苷能明显降低CCl4所致肝纤维化大鼠的肝脾指数(P<0.05)和血清中GPT,GOT活性(P<0.01),降低血清中HA,PCⅢ的含量(P<0.01);降低肝组织中MDA的水平和HyP的含量(P<0.01),亦能升高肝组织中SOD的活性(P<0.01)。结论:江苏地产白首乌总苷具有显著的抗肝纤维化的作用。
Objective: To study the protective effect of total glucosides of Polygonum multiflorum on liver fibrosis induced by carbon tetrachloride in rats. METHODS: Wistar rats were randomly divided into 6 groups: normal group, model group given 1% CMC-Na, positive drug group given hydrocortisone 3.5 mg.kg-1, Baishouwu low, middle and high dose groups were given white Shouwu total glycosides 4,8,16 mg.kg-1. Rats were given 5 mL of CCl4 peanut oil 2 mL.kg-1 twice weekly for 8 weeks. Modeling and administration are performed simultaneously. Eight weeks later, the blood was taken and the rat’s hepatic and spleen index was calculated. The activity of glutamic-pyruvic transaminase (GPT) and aspartate transaminase (GOT) in the serum of the rats was detected; the serum HA and procollagen III (PCIII) were detected. The content of superoxide dismutase (SOD), hydroxyproline (HyP) and malondialdehyde (MDA) in liver tissue were detected. Observe the pathological changes of liver tissue in rats. RESULTS: The total glucosides of Polygonum multiflorum in Jiangsu Province significantly reduced the liver and spleen index (P<0.05), serum GPT and GOT activity (P<0.01), and reduced the content of HA and PCIII in serum of rats with hepatic fibrosis induced by CCl4. (P<0.01); Decreasing the level of MDA and the content of HyP in liver tissue (P<0.01) also increased the activity of SOD in liver tissue (P<0.01). Conclusion: The total glucosides of Polygonum aviculare in Jiangsu Province has a significant anti-fibrotic effect.
