目的 筛选日本血吸虫感染者尿液检测探针。方法 采用电泳方法从日本血吸虫感染者尿液筛选诊断标志物,以此为抗原制备McAb,并用ELISA对尿样进行了初步检测。结果 PAGE电泳法筛选到一条分子量约30kDa 的糖蛋白带;以此糖蛋白为抗原免疫BALB/c 小鼠制备单克隆抗体,获得2 株McAb - NP56 和NP54;用NP56 以间接ELISA法检测10 份日本血吸虫感染者尿液(EPG中位数为69),浓缩50 倍尿液和原尿分别有9 份和5 份呈阳性。结论 本研究制备的McAb- NP56 有望作为日本血吸虫慢性病人和轻度感染者尿液的检测探针应用于现场。
Objective To screen the urine detection probe of Schistosoma japonicum infection. Methods Electrophoresis method was used to screen out the diagnostic markers from the urine of Schistosoma japonicum infected patients to prepare McAbs for antigens, and the urine samples were detected by ELISA. RESULTS: A glycoprotein band of about 30 kDa was screened by PAGE electrophoresis. BALB / c mice were immunized with the glycoprotein as antigen to prepare monoclonal antibodies. Two McAbs, NP56 and NP54, were obtained. Ten samples Urine of Schistosoma japonicum (EPG median of 69), urine 50% concentrated and original urine were 9 and 5 were positive. Conclusion McAb-NP56 prepared in this study is expected to be used as a detection probe for urine of chronic and mildly infected Schistosoma japonicum.
