目的分离纯化与富集山蓝中的红丝线素,并测定含量。方法山蓝全草经体积分数60%乙醇提取,D101大孔树脂层析色谱分离,制备型高效液相色谱制备红丝线素纯品,经HPLC测定含量,HPLC:采用Agilent柱,以甲醇-水(55∶45)为流动相,检测波长236 nm。结果红丝线素分离制备结果良好,在5.0~50.0 mg.L-1内线性良好(r=0.999 8),平均回收率97.67%,RSD为0.66%(n=9)。结论该方法可作为山蓝进一步研究的基础。
OBJECTIVE To isolate, purify and enrich red silk fibroin in mountain blue and determine its content. Methods The whole plant was extracted with 60% ethanol by volume fraction, separated by D101 macroporous resin chromatography and purified by preparative high performance liquid chromatography (HPLC). The content of HPLC was determined by HPLC. (55:45) as mobile phase, detection wavelength of 236 nm. Results The preparation of red silk fibroin was good. The average recovery was 97.67% and the RSD was 0.66% (n = 9) in the range of 5.0-50.0 mg · L-1 (r = 0.999 8). Conclusion This method can be used as the basis for the further research of Shanlan.
