在本实验条件下,EB与DNA及骨髓匀浆络合物的荧光光谱完全一致;标准DNA或RNA含量与荧光强度呈线性相关;DNA回收率为99.5％;方法灵敏度0.025μg/ml;最适pH范围5.5～7.7;离子强度范围0.073～0.292M;温度范围15～37℃。10只正常成年大鼠骨髓的DNA均值为15.2μg/mg骨髓,RNA为3.0μg/mg骨髓。此法亦可测定白细胞DNA含量。
Under the experimental conditions, the fluorescence spectra of EB and DNA and bone marrow homogenate complexes were completely consistent; the standard DNA or RNA content was linearly correlated with the fluorescence intensity; the DNA recovery was 99.5%; the sensitivity was 0.025μg / ml; pH range 5.5 ~ 7.7; ionic strength range 0.073 ~ 0.292M; temperature range 15 ~ 37 ℃. The mean DNA value of bone marrow from 10 normal adult rats was 15.2 μg / mg bone marrow with RNA at 3.0 μg / mg bone marrow. This method can also determine the leukocyte DNA content.
