以30份甜瓜种质资源为材料,采用RAPD(random amplified polymorphic DNA)、DAMD(directed amplification of minisatellite-region DNA)和SRAP(sequence-related amplified polymorphism)3种分子标记构建各自的DNA指纹图谱。针对图谱上扩增的条带分子量大小和分布,用统计软件逐一聚类,根据不同引物(引物组合)所得聚类图鉴别的品种数目,筛选出高分辨率的4个RAPD引物、3个DAMD引物和6个SRAP引物组合。同时,利用高效引物(引物组合)评价了甜瓜遗传多样性,将30份甜瓜分为薄皮和厚皮两类,厚皮甜瓜包括网纹和无网纹两种类型。
Thirty kinds of melon germplasm resources were used as materials to construct their DNA fingerprints using random amplified polymorphic DNA (RAPD), directed amplification of minisatellite-region DNA (DAMD) and sequence-related amplified polymorphism (SRAP). According to the size and distribution of bands amplified on the map, one by one using the statistical software to cluster, according to the number of varieties identified by different primer (primer combination) clustering map to identify high-resolution 4 RAPD primers, 3 DAMD Primer and 6 SRAP primer combinations. At the same time, the genetic diversity of melon was evaluated by high-performance primer (primer combination). The melon was divided into two types, thin-skinned and thick-skinned, including muskmelon and anilox.
