Glypican-3(GPC3)在肝细胞癌(肝癌)中过表达,而在成人正常组织、癌旁组织、肝炎、肝硬化组织中仅低表达或不表达。GPC3mRNA在小肝癌检出率明显比甲胎蛋白(AFP)mRNA和血清中的AFP高。以上研究是用northernblot、原位分子杂交等技术检测GPC3mRNA,本研究用逆转录聚合酶链反应(RT-PCR)方法检测肝癌及癌旁组织中GPC3mRNA的表达情况,探讨GPC3在肝癌发生发展中的作用。一、材料与方法1.材料:标本的收集:原发性肝癌及其相应的癌旁组织
Glypican-3 (GPC3) is overexpressed in hepatocellular carcinoma (HCC) and only lowly or not expressed in adult normal tissues, paracancerous tissues, hepatitis and cirrhosis tissues. The detection rate of GPC3 mRNA in small hepatocellular carcinoma was significantly higher than that in AFP mRNA and serum. In the above study, GPC3 mRNA was detected by northernblot and in situ hybridization. In this study, GPC3 mRNA expression in hepatocellular carcinoma and its adjacent tissues was detected by reverse transcription-polymerase chain reaction (RT-PCR), and GPC3 mRNA expression in hepatocellular carcinoma effect.
