目的:了解EB病毒编码BHRF1基因表达对鼻咽癌细胞增殖周期的影响。方法:本研究通过构建BHRF1高表达载体并将其转入鼻咽癌细胞株CNE2细胞中,检测转染后稳定表达BHRF1癌细胞的增殖细胞核抗原(proliferatingcelnuclearantigen,PCNA)表达和喜树碱作用前后细胞周期分布的改变。结果:BHRF1的表达可促使癌细胞的PCNA表达下降,G1期细胞增多,S期细胞减少;而在喜树碱作用后,BHRF1-CNE2细胞的凋亡率明显小于对照组,其G1期细胞数下降,S期细胞数升高。结论:BHRF1基因的表达可通过调节细胞周期分布、抑制细胞凋亡的发生以增强细胞的生存能力,而可能与鼻咽癌的发生有关
OBJECTIVE: To investigate the effect of EBR-encoded BHRF1 gene expression on the proliferation of nasopharyngeal carcinoma cells. Methods: In this study, BHRF1 high expression vector was constructed and transfected into CNE2 cells. The expression of proliferating cell nuclear antigen (PCNA) and the expression of PCNA in BHRF1 cells after transfection were detected. Changes in the periodic distribution. Results: The expression of BHRF1 decreased the expression of PCNA in the cancer cells, but increased in the G1 phase and decreased in the S phase. However, the apoptosis rate of BHRF1-CNE2 cells was significantly lower than that of the control group after the camptothecin treatment. The number of G1 phase cells Declining, S-phase cells increased. Conclusion: The expression of BHRF1 gene may be related to the occurrence of nasopharyngeal carcinoma by regulating cell cycle distribution and inhibiting the occurrence of apoptosis in order to enhance cell viability
