目的采用碱水冷浸法从槐花米中提取槲皮素,并优化制备工艺。方法以星点设计效应面法设计优化试验,以料液比,提取时间,提取pH值为考察因素,以产量和纯度为指标,分别采用多元线性回归和二项式方程拟合实验结果,绘制效应面和等高线图,由此优化工艺参数。结果槐花米中槲皮素提取的最佳条件为:料液比35,pH 10,提取时间为24h。结论该工艺简单易行,未使用有机溶剂,能耗低,可为工业化生产提供参考。
OBJECTIVE To extract quercetin from Sophora japonica by alkaline water-cooling method and to optimize the preparation process. Methods The optimization design was carried out by the design of response surface methodology with star point design. The ratio of solid to liquid, extraction time and pH value were taken as the investigation factors. The yield and purity were used as indices. Multivariate linear regression and binomial equation were used to fit the experimental results respectively. Effect surface and contour map, thereby optimizing the process parameters. Results The optimum conditions for the extraction of quercetin from Sophora japonica L. were as follows: the ratio of material to liquid was 35, the pH was 10 and the extraction time was 24h. Conclusion The process is simple and easy, without the use of organic solvents, low energy consumption, which can provide reference for industrial production.
