目的研究卵巢摘除对于急性肝损伤模型鼠脑血管内皮细胞功能的影响。方法对去势(卵巢摘除)与未去势小鼠分别给与四氯化碳制备急性肝损伤模型,另设单纯卵巢摘除组及空白对照组;免疫组织化学法观察各组小鼠脑血管内皮细胞Bax及Bcl-2的表达。结果发现各组小鼠嗅脑及皮层等脑区毛细血管内皮细胞Bax阳性表达结果类似;而Bcl-2表达存在明显差异,急性肝损伤组嗅脑及皮层等各脑区毛细血管内皮细胞Bcl-2阳性表达呈强阳性;而去势+急性肝损伤组各脑区Bcl-2的阳性表达均偏弱。结论卵巢摘除或卵巢功能退化会影响脑血管内皮细胞应对化学毒性损伤时的抗凋亡功能,从另一个侧面揭示了卵巢相关激素的脑保护作用。
Objective To study the effect of ovariectomy on the function of cerebrovascular endothelial cells in acute liver injury model rats. Methods Acute liver injury model was induced by castrated carbon tetrachloride (CCl4) in ovariectomized (OVX) and untargeted mice. A simple ovariectomized group and a blank control group were also established. Immunohistochemistry was used to observe the changes of cerebral vascular endothelium Cell Bax and Bcl-2 expression. The results showed that Bax positive expression of capillary endothelial cells in the brain regions such as olfactory brain and cortex of mice in each group were similar, but the expression of Bcl-2 was significantly different in each group. The expression of Bcl- 2 positive expression was strong positive; and castrated + acute liver injury group brain area Bcl-2 positive expression is weaker. Conclusion Degeneration of ovary or ovary may affect the anti-apoptotic function of cerebrovascular endothelial cells in response to chemical toxic injury, revealing the protective effect of ovarian related hormones from another aspect.
