目的观察不同浓度羊自体血清对骨髓间充质干细胞(bone marrow-derived mesenchymal stem cells,bMSCs)增殖活力的影响,并筛选出最佳血清培养浓度。方法以12月龄青山羊为实验动物,首先制备自体血清,分离获取bMSCs;然后选DMEM/F12为基础培养基,采用不同体积分数(分别为5%、10%、15%)自体血清及10%体积分数胎牛血清培养基分别对bMSCs进行培养;通过显微镜下细胞形态学观察、活细胞计数及MTT等方法对细胞进行检测,并对检测数据做统计学分析。结果不同浓度自体血清培养培养条件下bMSCs形态正常。与10%胎牛血清组进行比较,5%自体血清组bMSCs增殖较慢(P<0.01),10%、15%自体血清组增殖较好,但组间无统计学差异(P>0.05)。结论羊自体血清培养bMSCs是可行的,10%浓度自体血清是最佳培养浓度。
Objective To observe the effects of different concentrations of goat serum on the proliferation of bone marrow-derived mesenchymal stem cells (bMSCs) and to screen out the best serum culture concentration. Methods 12-month-old young goats were used as experimental animals. First, autologous serum was prepared and bMSCs were isolated. Then, DMEM / F12 was used as the basal medium. Different concentrations of serum (5%, 10%, 15% The bMSCs were cultured in% FCM. The cells were detected by microscope, viable cell count and MTT. The detection data were analyzed statistically. Results Different concentrations of autologous serum culture bMSCs under normal culture conditions. Compared with 10% fetal bovine serum group, 5% autologous serum group bMSCs proliferated slowly (P <0.01), and 10%, 15% autologous serum group proliferated well, but there was no significant difference between the two groups (P> 0.05). Conclusion It is feasible to culture bMSCs with sheep autologous serum and 10% concentration of autologous serum is the best culture concentration.
