目的采用制备色谱系统从九节龙皂苷粗品中分离纯化九节龙皂苷Ⅰ,建立其纯化工艺。方法九节龙皂苷粗品采用柱色谱分离,然后采用制备色谱系统,收集对应峰的回收液,即得九节龙皂苷Ⅰ,并对产品进行了TLC定性和质量分数定量分析。结果产品经HPLC检测,为九节龙皂苷Ⅰ,质量分数达99%以上。结论用制备色谱系统进行九节龙皂苷Ⅰ的分离和纯化,具有简便、快捷、周期短、自动化程度高、收集全面等优点。
Objective To separate and purify N. japonicum saponin I from the crude product of N. japonicum with preparative chromatographic system and establish its purification process. Methods The crude product of Nine isoflavones was separated by column chromatography. Then the prepared chromatographic system was used to collect the recovery solution of the corresponding peaks. The N. japonicus saponin I was obtained, and the product was quantitatively analyzed by qualitative TLC and mass fraction. Results The product was detected by HPLC and was Nine Diosaponin I with a mass fraction of more than 99%. Conclusion Separation and purification of N. japonicum I with a preparative chromatographic system has the advantages of simplicity, rapidity, short cycle, high degree of automation, and comprehensive collection.
