目的 :在单纯性肥胖大鼠中研究其肥胖特点及瘦素的变化。方法 :6 0只新生SD大鼠随机分成两组 ,肥胖组 (n =40 )喂高能量饲料 ,对照组 (n =2 0 )喂普通饲料 ,动态观察体重、腹腔脂肪的变化 ,用免疫组化(ABC法 )法观察腹腔脂肪细胞瘦素表达 ,用放免法测血清瘦素水平。结果 :肥胖组大鼠与非肥胖对照组大鼠比 ,第 7周体重增长最显 ,12周内体重持续上升 ,12周时腹腔脂肪重量增加 ,脂肪细胞增大 ,脂肪细胞瘦素表达增强 ,且与腹腔脂肪组织重量呈明显正相关 ,肥胖大鼠血清瘦素水平明显低于对照组 (P <0 .0 5 )。结论 :在单纯性肥胖大鼠中体重、Lee’s指数、腹腔脂肪重量、脂肪细胞大小变化显著。肥胖鼠的脂肪细胞瘦素表达增强 ,并与腹腔脂肪重量呈正相关 ,因此它可作为衡量大鼠肥胖程度的一项指标
OBJECTIVE: To study the obesity characteristics and leptin changes in simple obese rats. Methods: Sixty newborn SD rats were randomly divided into two groups. The rats in the obesity group (n = 40) were fed with high-energy diet and the control group (n = 20) fed the normal diet. The body weight and abdominal fat were dynamically observed. (ABC method) was used to observe the expression of leptin in peritoneal adipocytes, serum leptin level was measured by radioimmunoassay. Results: Compared with the non-obese rats, the body weight gain was the highest in the 7th week in obese rats and the body weight increased continuously in 12 weeks. The weights of abdominal fat increased, the fat cells increased, the leptin increased in adipocytes, (P <0.05). The level of serum leptin in obese rats was significantly lower than that in control rats (P <0.05). Conclusion: Body weight, Lee’s index, weight of abdominal fat and fat cell size in simple obese rats changed significantly. Lipid expression in adipocytes of obese rats is enhanced and positively correlated with abdominal fat weight, so it can be used as a measure of obesity in rats
