【目的】探讨补肾活血汤对骨髓间充质干细胞(BMSCs)迁移及趋化因子受体CXCR4表达的影响。【方法】以大鼠BMSCs为研究对象,采用全骨髓贴壁法培养BMSCs。采用流式细胞术鉴定细胞表面抗原CD90、CD34、CD11b;采用Transwell试验检测不同剂量组补肾活血汤含药血清对BMSCs迁移的影响;采用Western blot法检测细胞CXCR4的蛋白表达水平。【结果】细胞表面抗原标记结果显示所得细胞为高纯度BMSCs。Transwell结果显示,补肾活血汤含药血清可显著增强大鼠BMSCs的迁移能力;Western blot结果显示,高、中、低剂量组补肾活血汤含药血清呈浓度依赖性上调大鼠BMSCs的CXCR4蛋白表达,与空白血清组比较,差异均有统计学意义(P<0.05)。【结论】补肾活血汤含药血清可促进BMSCs的体外迁移,并影响CXCR4蛋白的表达,其机制可能与上调CXCR4的表达、激活SDF-1/CXCR4轴有关。
【Objective】 To investigate the effects of Bushen Huoxue Decoction on the migration of bone marrow mesenchymal stem cells (BMSCs) and the expression of chemokine receptor CXCR4. 【Method】 BMSCs were cultured in whole bone marrow using rat BMSCs as the research object. The cell surface antigen CD90, CD34 and CD11b were identified by flow cytometry. Transwell assay was used to detect the effects of different doses of Bushen Huoxue decoction on the migration of BMSCs. The protein expression of CXCR4 was detected by Western blot. 【Result】 The results of cell surface antigen labeling showed that the obtained cells were high purity BMSCs. Transwell results showed that Bushenhuoxue Decoction-containing serum could significantly enhance the migration of BMSCs in rats; Western blot results showed that Bushenhuoxue Decoction-containing serum in a dose-dependent manner upregulated CXCR4 protein expression in BMSCs of rats , Compared with the blank serum group, the difference was statistically significant (P <0.05). 【Conclusion】 Bushen Huoxue Decoction containing serum can promote the migration of BMSCs in vitro and affect the expression of CXCR4 protein, which may be related to the up-regulation of CXCR4 expression and the activation of SDF-1 / CXCR4 axis.
