目的:研究牛蒡苷胶囊对阿霉素肾病大鼠的保护作用。方法:采用一次性尾静脉注射(阿霉素ADR)复制微小病变型肾病模型,。SD雄性大鼠随机分为空白组、模型组、阳性药醋酸泼尼松片5 mg.kg-1组、牛蒡苷胶囊高、中、低剂量组(767,383,192 mg.kg-1),ig给药6周。于每周末将大鼠放入代谢笼中,收集24 h尿液,用全自动生化仪测定尿蛋白含量。于末次给药后24 h,眼眶静脉丛采血,于全自动生化仪测定白蛋白(ALB)、甘油三酯(TG)、胆固醇(TC)含量;采用酶联免疫法测定血清转化生长因子-β1(TGF-β1)水平;采用黄嘌呤氧化酶法和硫代巴比妥法分别测定血清超氧化物歧化酶(SOD)和丙二醛(MDA);肾组织病理学检测。结果:与模型组比较,牛蒡苷胶囊高、中剂量能减少阿霉素肾病大鼠的尿蛋白含量,提高血清ALB含量;牛蒡苷胶囊3个剂量组能降低血清TG和TGF-β1含量;牛蒡苷高剂量(767 mg.kg-1)能提高血清SOD活性、降低血清MDA含量;牛蒡苷胶囊3个剂量组能够促进肾组织病理改变的修复。结论:牛蒡苷胶囊对阿霉素肾病大鼠的保护作用可能与其清除氧自由基、抑制脂质过氧化反应、抑制肾小球纤维化等作用有关。
Objective: To study the protective effect of arctiin capsule on adriamycin-induced nephropathy in rats. Methods: One-time tail vein injection (Adriamycin ADR) was used to replicate the minimal change nephropathy model. SD male rats were randomly divided into blank group, model group, positive prednisone acetate tablets 5 mg.kg-1 group, burdock capsule high, medium and low dose groups (767,383,192 mg.kg-1), ig administration 6 weeks. Rats were placed in metabolic cages every weekend, urine samples were collected for 24 hours, and urine protein content was determined by automatic biochemical analyzer. At 24 h after the last administration, orbital venous plexus blood was collected and the content of albumin (ALB), triglyceride (TG) and cholesterol (TC) were measured by automatic biochemical analyzer. The levels of serum transforming growth factor-β1 (TGF-β1). Serum levels of superoxide dismutase (SOD) and malondialdehyde (MDA) were measured by xanthine oxidase method and thiobarbiturine method respectively. Results: Compared with the model group, high and middle dose of burdock capsule could decrease the urinary protein content and increase the content of ALB in the adriamycin-induced nephropathy rats. The three doses of the burdock capsule could decrease the contents of TG and TGF-β1 in serum. High dose of glycosides (767 mg.kg-1) increased serum SOD activity and decreased serum MDA content. Arctiin capsule 3-dose group could promote the repair of renal pathological changes. Conclusion: The protective effect of arctiin capsule on adriamycin-induced nephropathy rats may be related to its function of scavenging oxygen free radicals, inhibiting lipid peroxidation and inhibiting glomerular fibrosis.
